技術簡介
工研院成功製備出世界上第一個具有可控制多突狀型態的微米級磁珠,稱為iKNOBeads,其均勻的尺寸及特有的多突狀型態提供優異的再現性與反應性,並可藉由表面修飾物質的變化應用於CAR-T及NK細胞的生產,基於iKNOBeads所建立的免疫細胞生產製程,經臨床檢體驗證能夠產出量多質優且具有高毒殺能力的細胞,效能超越國際領導廠商競品,同時也已建立磁珠GMP製程並獲得衛福部核可登錄(QMS 1850)。
Abstract
ITRI has invented the world’s first micrometer-sized magnetic beads, named iKNOBeads, with tailorable sizes and shapes. Its uniform size and unique bumpy morphology provide excellent reproducibility and reactivity toward immune cells. We have demonstrated that manufacturing CAR-T and NK cells can be done through changes in surface stimulatory ligands. In a healthy-donor and cancer-patient study, immune cells’ proliferation capability, quality, and cytotoxicity expanded with iKNOBeads are superior to commercially available products. These results show that iKNOBeads offers a more effective and economical feeder-free platform for generating high-quality and quantity immune cells for therapy. GMP-compliant

Technical Specification
Lipid nanoparticles (size between 50-200 nm depending on formulations, Polydisperdisity inex <0.2 , encapsulation efficiency 70-99%). Candidate lipid materials and formulations have been assessed for in vivo distribution and show in vivo protein expression efficacy comparable or superior to commercial formulations.
