技術名稱: 光學活性物合成

技術簡介

不對稱合成及酵母菌還原法為原料藥開發不可或缺的技術,利用此技術建立具光學活性C4關鍵中間體(3R)-3-hydroxy-4-halo-butanoate之實驗室製程:方法一:利用不對稱合成,分別合成6種不對稱-RuLXQ(L=S-binap or S-T-binap;X=C1 or OAc;Q=dmf or Et3N),分別在30-110℃/5-100atm下進行研究,同時亦針對觸媒重複使用、添加劑對反應性及光學活性影響進行探討,產物經由簡單蒸餾穫得。方法二:以酵母菌還原法,分別就baker yeast用量、基質濃度(1-10%)、探討反應性及光學活性影響此二種方法均可得C4關鍵中間體(3R)-3-hydroxy-4-halo-butanoate,產物光學純度(enantiomeric excess值)>95%、產品純度(purity)>95%、產率>85%。
