技術名稱: 藥材DNA鑑定技術及300種常用中藥材ITS鑑定序列資料庫

技術簡介

本技術是關於利用DNA技術鑑定中藥材及300種常用中藥材ITS鑑定序列資料庫,可提供作為比對鑑定的依據。本技術係根據植物染色體DNA多型性(polymorphism)的特性,在DNA層次上鑑定與管控藥材基原。可使用的分子技術很多,但因藥材多經曝曬及炮製,DNA取得不易,在簡單、快速、符合經濟效益及準確的考量下,以最靈敏的聚合脢鏈反應的技術複製藥材DNA ,結合核醣體基因間(internal transcribed spacer,ITS)序列差異的比對技術,僅需要少量的藥材樣品就可以快速準確的鑑定出形態類似的藥材以作為鑑定依據。目前已建構300種常用中藥材的ITS鑑定序列資料庫,可作為中草藥業界或物流廠商掌控品管藥材的依據。相關技術包括藥材DNA萃取技術、特殊引子設計、ITS片段複製及序列解讀比對技術,是未來中草藥產品通過國內衛生署及美國FDA核准時正確基原最佳的參考依據。

Abstract

This technology is describing development of DNA technology to identify Chinese herbal materials and database construction of internal transcribed spacer (ITS) sequences of 300 common-used Chinese herbal materials based on the DNA polymorphism occurred in chromosomes. Considering the difficulty in obtaining intact DNA from herbal materials, the polymerase chain reaction was conducted and the region of internal transcribed spacers of rRNA genes was amplified and sequenced. Only small amounts of DNA will meet the need, which is suitable for identification of Chinese herbal materials that have been processed and exposed in the sun or stored for a long time. The related techniques include DNA isolation from Chinese herbal materials, specific primer design, amplification of ITS region, and comparison of obtained sequences to clarify the materials.We could foresee that the industrial sectors could apply and promote this database to authenticate Chinese herbal materials for decreasing misuse and ensuring quality, moreover, for being as the base of applying FDA or DOH approval.
