技術簡介

該技術係先固定小鼠抗體之重鏈或輕鏈，配合可變之人類輕鏈或重鏈基因組，針對特定標靶蛋白篩選得到最適之人類輕鏈或重鏈結合者後，再固定之，並將另一鏈改為可變的重鏈或輕鏈，用相同方法篩選，最後可得到一完全擬人化的抗體

Abstract

This is an antibody humanization technique which involves retaining murine heavy/light chain variable region and shuffling the light/heavy chain variable region with human antibody library. After obtaining the best binder by phage display, retain the binder and shuffle the other chain. As a result, a human new antibody is selected out.

應用範圍

作為快速篩選有潛力之診斷或治療用擬人化抗體之用，進而得到高專一性和高親和力的抗體，並降低在人體內被分解以及引發免疫反應 (HAMA, human anti-mouse antibody response)的風險。
