技術名稱: 熱對流式核酸擴增即時檢測方法及裝置

技術簡介

以熱對流聚合?連鎖反應(convective PCR, cPCR)原理,具有即時檢測功能之Real-time ccPCR (capillary convective PCR)雛型系統,並具有即時定量偵測功能,包含光學模組設計、機構設計、電控模組設計、操作介面設計、以及反應試劑設計,並呈現系統與試劑之可行性測試。

Abstract

In view of the potential applications of thermal convection polymerase chain reaction (convective PCR, cPCR), as well as the current unmet needs of exclusive real-time detection function, the goal of this study is to develop a real-time capillary convection PCR(ccPCR) prototype system, which is capable of real-time and quantitative detection. In this report, it contains the detail about optical module design, mechanical design, electronic control module design, interface design, and reagents design. Feasibility tests of both the prototype system and reagents were demonstrated in the end.

技術特色

*技術現況: 熱對流式PCR技術最早是由Krishnan等人設計了Rayleigh-Benard cell的圓柱形腔體,配合上下兩個不同加熱源;之後還有封閉迴路式流道,配合三種加熱源;或非接觸輻射僅在中央加熱式的封閉腔體。目前的市售系統僅提供加熱器的功能、或只容許在PCR反應結束時做定性判別(positive或negative),尚未建立即時定量偵測(quantitative real-time detection)的能力。 *本團隊研發特色: 1.以熱對流式PCR技術(convective PCR, cPCR)為基礎之小型化PCR反應裝置,整體為可移動式,取代目前市場上的大型熱傳導式thermal cycler. 2.PCR反應放大時間: < 30分鐘,樣本不需要特別預熱。 3.僅需要單一加熱源。 4.能以光偵測器偵測PCR產物隨時間的變化量。 5.能同時分析多管的反應變化量。 6.簡易操作之人機介面系統。 7.反應容器搭配溫控模組,使內部溫度梯度滿足PCR試劑要求的反應溫度區間。

應用範圍

此小型化、可移動式之核酸擴增即時檢測系統,適用於各醫護點使用,可結合試劑開發發展各類分子診斷應用,如:致病原檢測及標的基因檢測,及發展為更簡便之居家用核酸分子檢測潛力。
